Czy jednak jest to w\u0142a\u015bciwy trop? Hodowla in vitro zarodk\u00f3w, mimo i\u017c najbardziej znana, to tylko wierzcho\u0142ek g\u00f3ry lodowej zastosowa\u0144 tej techniki.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"co-to-in-vitro\">Co to in vitro?<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">\u201eIn vitro\u201d po \u0142acinie oznacza \u201ew szkle\u201d. <a id=\"_ednref1\" href=\"#_edn1\">[i]<\/a> Badania in vitro to zatem nic innego jak przeniesienie reakcji zachodz\u0105cych normalnie w \u017cywym organizmie do warunk\u00f3w laboratoryjnych. Dotyczy to zar\u00f3wno reakcji chemicznych, jak r\u00f3wnie\u017c wzrostu i r\u00f3\u017cnicowania kom\u00f3rek i tkanek.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"dlaczego-in-vitro\">Dlaczego in vitro?<\/h2>\n\n\n\n<div class=\"wp-block-columns is-layout-flex wp-container-core-columns-is-layout-8f761849 wp-block-columns-is-layout-flex\">\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\">\n<p class=\"wp-block-paragraph\">Mo\u017cna powiedzie\u0107, \u017ce pierwsze eksperymenty polegaj\u0105ce na obserwowaniu kom\u00f3rek nerwowych \u017caby okaza\u0142y si\u0119 prawdziwym prze\u0142omem [2]. Mimo i\u017c R. G. Harrison nie zna\u0142 sposobu na utrzymanie hodowli przy \u017cyciu, da\u0142 pocz\u0105tek rozwoju niesamowitego narz\u0119dzia poznawczego dla nauki i medycyny. Dzi\u0119ki odzwierciedleniu warunk\u00f3w panuj\u0105cych w \u017cywym organizmie, mo\u017cna bada\u0107 procesy, kt\u00f3rych nie da si\u0119 zasymulowa\u0107 na sztucznych modelach. Hodowle kom\u00f3rek i tkanek in vitro uniezale\u017cni\u0142y ponadto w du\u017cym stopniu badaczy od eksperymentowania na zwierz\u0119tach. Pozwoli\u0142o to w wielu przypadkach wyeliminowa\u0107 problem etyczny. Obecnie trudno przeceni\u0107 rol\u0119 hodowli in vitro w rozwoju nauki, medycynie, biotechnologii. Zastosowanie hodowli kom\u00f3rek i tkanek jest powszechne i wykorzystywane r\u00f3wnie\u017c na skal\u0119 przemys\u0142ow\u0105. U\u017cywa si\u0119 ich mi\u0119dzy innymi do bada\u0144 cytotoksyczno\u015bci substancji chemicznych. Dzi\u0119ki modelom imituj\u0105cym kosmki jelitowe bada si\u0119 przepuszczalno\u015b\u0107 jelita dla lek\u00f3w oraz innych zwi\u0105zk\u00f3w. Kom\u00f3rki w hodowli in vitro produkuj\u0105 bia\u0142ka, np. insulin\u0119 na skal\u0119 przemys\u0142ow\u0105. Mo\u017cna wyhodowa\u0107 fragment sk\u00f3ry i u\u017cy\u0107 go do autoprzeszczepu. Po pierwszym pasa\u017cu hodowla pierwotna staje si\u0119 hodowl\u0105 wt\u00f3rn\u0105. Najcz\u0119\u015bciej kom\u00f3rki w hodowli pierwotnej przyczepiaj\u0105 si\u0119 do pod\u0142o\u017ca tworz\u0105c pojedyncz\u0105 warstw\u0119.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Hodowla in vitro w zawiesinie<\/strong> mog\u0105 by\u0107 prowadzone dla kom\u00f3rek, kt\u00f3re mog\u0105 rosn\u0105\u0107 i proliferowa\u0107 niezale\u017cnie od przyczepiania si\u0119 do pod\u0142o\u017ca. Dotyczy to m.in. linii kom\u00f3rkowych, kom\u00f3rek nowotworowych lub kom\u00f3rek krwi.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Hodowle narz\u0105dowe <\/strong>pozwalaj\u0105 najlepiej odtworzy\u0107 sytuacj\u0119 in vivo. Dzi\u0119ki hodowli ca\u0142ych narz\u0105d\u00f3w lub ich fragment\u00f3w zostaj\u0105 zachowane naturalne interakcje mi\u0119dzy kom\u00f3rkami, co pozwala na uzyskanie wysoce wiarygodnych wynik\u00f3w.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Hodowle na mikrono\u015bnikach<\/strong> daj\u0105 mo\u017cliwo\u015b\u0107 wzrostu du\u017cej ilo\u015bci kom\u00f3rek adherentnych w stosunkowo niewielkiej obj\u0119to\u015bci medium. S\u0105 to hodowle prowadzone na kulkach wykonanych z r\u00f3\u017cnego rodzaju tworzywa (dekstranowych, szklanych, \u017celatynowych, plastikowych).<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Hodowle przestrzenne<\/strong> prowadzone s\u0105 w celu stworzenia modeli na\u015bladuj\u0105cych tkanki i narz\u0105dy \u017cywego organizmu. Zalicza si\u0119 do nich hodowle narz\u0105dowe, agregaty i sferoidy, kokultury organotypowe, hodowle na sztucznych naczyniach w\u0142osowatych, modele tkankowe i narz\u0105dowe.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"czy-hodowle-komorkowe-i-tkankowe-wymagaja-specjalnych-warunkow\">Czy hodowle kom\u00f3rkowe i tkankowe wymagaj\u0105 specjalnych warunk\u00f3w?<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Wyposa\u017cenie pracowni in vitro wymaga odpowiedniej wiedzy i przygotowania. Hodowle musz\u0105 odbywa\u0107 si\u0119 w sterylnych warunkach z zachowaniem bardzo restrykcyjnych zasad aseptyki. Nale\u017cy dezynfekowa\u0107 sprz\u0119t laboratoryjny oraz powierzchnie. Ponadto naczynia hodowlane mo\u017cna otwiera\u0107 wy\u0142\u0105cznie w komorze laminarnej. Hodowle in vitro prowadzone s\u0105 w okre\u015blonych warunkach wilgotno\u015bci, temperatury i zawarto\u015bci CO<sub>2<\/sub>. U\u017cywa si\u0119 medium hodowlanego z zawarto\u015bci\u0105 substancji od\u017cywczych i soli mineralnych. Ma to na celu w jak najwi\u0119kszym stopniu upodobni\u0107 warunki hodowli do tych panuj\u0105cych w \u017cywym organizmie.
